< Abstract >

This study was designed to evaluate the effect of a gel formula containing porcine placenta extract (KX-2) on skin and its mechanism. The treatment of KX-2 increased stratum corneum (SC) water contents, and alleviated wrinkle index significantly after 6-weel treatment with KX-2 gel in vivo, while KX-2 did not affected skin elasticity. Next, we evaluated extracellular matrix production. The productions of collagen and hyaluronan (HA) from normal human dermal fibroblasts were not significantly affected by KX-2. Further, hyaluronan production from human epidermal keratinocytes was not affected. These results are consistent with there was no significant change in skin elasticity. Additionally, Theses suggest that the increase in SC water contents was not due to HA. On the other hand, the expression of CERS3, a major ceramide synthase in epidermis, was significantly enhanced by KX-2. Taken together, the results suggest that alleviation of wrinkle index was caused by increase in SC water content upon enhanced CERS3 expression.
